{"id":463,"date":"2020-12-06T22:22:48","date_gmt":"2020-12-06T22:22:48","guid":{"rendered":"http:\/\/www.apus.uno\/noticias\/?p=463"},"modified":"2020-12-06T22:24:46","modified_gmt":"2020-12-06T22:24:46","slug":"enfermedades-geneticas","status":"publish","type":"post","link":"https:\/\/www.apus.uno\/noticias\/2020\/12\/06\/enfermedades-geneticas\/","title":{"rendered":"CURA DEFINITIVA PARA LAS ENFERMEDADES GEN\u00c9TICAS"},"content":{"rendered":"\n<p>El editor de ADN <strong>\u201cPRIME EDITING\u201d&nbsp; <\/strong>podr\u00eda curar casi el 90% de las enfermedades gen\u00e9ticas<\/p>\n\n\n\n<p>Una nueva t\u00e9cnica denominada \u201cPRIME EDITING\u201d &nbsp;Corrector ortogr\u00e1fico del Genoma, podr\u00eda evitar el 89% de las cerca de 75.000 enfermedades gen\u00e9ticas conocidas, en un tiempo y con precisi\u00f3n r\u00e9cord.<\/p>\n\n\n\n<p>Descubierta por el considerado como uno de los mejores cient\u00edficos del planeta, el qu\u00edmico estadounidense David Liu, de la Universidad de Harvard.<\/p>\n\n\n\n<p>Las c\u00e9lulas humanas tienen su manual de instrucciones escrito en dos metros de ADN plegados de manera asombrosa, en el lenguaje ARN.<\/p>\n\n\n\n<p>En la t\u00e9cnica anterior y utilizada por los laboratorios desde el 2.012, el CRIPSPR- Cas9, era capaz de buscar una secuencia concreta de letras del ARN y cortarla con una especie de tijeras moleculares y pegar fragmentos del ADN, insertando nueva informaci\u00f3n sintetizada por los cient\u00edficos como si fuera un procesador de textos.<\/p>\n\n\n\n<p>El problema es que, a menudo, la operaci\u00f3n falla y se generan mutaciones no deseadas.<\/p>\n\n\n\n<p>El a\u00f1o pasado nacieron en China&nbsp;<a href=\"https:\/\/elpais.com\/elpais\/2019\/03\/01\/ciencia\/1551430968_283321.html\">los dos primeros beb\u00e9s cuyo genoma fue modificado<\/a>&nbsp; (t\u00e9cnica CRISPR) para que fuesen inmunes al virus del sida. Este avance logrado por el cient\u00edfico He Jiankui, recibido con alarma por la comunidad cient\u00edfica porque la t\u00e9cnica puede generar mutaciones no deseadas en otras partes del genoma.<\/p>\n\n\n\n<p>La t\u00e9cnica descubierta por David Liu, y bautizada como PRIME EDITING (\u201cedici\u00f3n de calidad\u201d) sirve para reparar el 89% de las 75.000 variantes gen\u00e9ticas humanas asociadas a enfermedades no se pueden corregir actualmente en el laboratorio.<\/p>\n\n\n\n<p>A diferencia del CRISPR, que son unas tijeras que rompen la doble cadena del ADN, el editor \u201cPRIME EDITING\u201d es un sistema de &#8216;b\u00fasqueda y sustituci\u00f3n&#8217; que permite realizar directamente mutaciones puntuales espec\u00edficas, inserciones y eliminaciones de una sola letra precisas y combinaciones de estas, sin tener que romper la doble cadena.<\/p>\n\n\n\n<p>La estrategia de David Liu es diferente, ya que ha&nbsp;<em>inventado<\/em>&nbsp;\u201cuna nueva prote\u00edna quim\u00e9rica\u201d, capaz de evita as\u00ed las mutaciones indeseadas que se produc\u00edan con &nbsp;las tijeras Cas9.<\/p>\n\n\n\n<p>Para dirigir su m\u00e1quina molecular a un lugar concreto del genoma para escribir una nueva informaci\u00f3n&nbsp; gen\u00e9tica directamente en el genoma. Liu utiliza una gu\u00eda de ARN y \u201cuna transcriptasa reversa\u201d una prote\u00edna que usan los virus para copiar su ARN en ADN, invirtiendo el flujo can\u00f3nico de la informaci\u00f3n gen\u00e9tica, que parte del ADN y se copia a ARN, para acabar convirti\u00e9ndose en una prote\u00edna\u201d.<\/p>\n\n\n\n<p>Seg\u00fan el genetista y cient\u00edfico del Centro Nacional de Biotecnolog\u00eda (CNB) Llu\u00eds Montoliu, se muestra&nbsp;<strong>entusiasmado con este \u00abcambio revolucionario\u201d<\/strong> \u201cLa gu\u00eda de ARN en este caso se extiende y tiene un extremo nuevo, m\u00e1s largo, que es usado como molde por la transcriptasa reversa para copiar nuevo ADN con la secuencia correcta, con la mutaci\u00f3n corregida\u201d.<\/p>\n\n\n\n<p>El equipo de Liu ha demostrado en l\u00edneas celulares de laboratorio que el&nbsp;<em>prime editing<\/em>&nbsp;genera menos errores de edici\u00f3n en los lugares a los que va dirigido, aunque no ha analizado si hay errores fuera de sitio.<\/p>\n\n\n\n<p>Adem\u00e1s a publicado&nbsp; <a href=\"https:\/\/www.nature.com\/articles\/s41586-019-1711-4\">en la revista&nbsp;<em>Nature<\/em><\/a>&nbsp;los resultados de 175 experimentos en c\u00e9lulas humanas en el laboratorio, incluyendo la correcci\u00f3n de las causas gen\u00e9ticas de trastornos como la anemia de c\u00e9lulas falciformes y la enfermedad de Tay-Sachs.<\/p>\n\n\n\n<p>Hilary Sheppard, bi\u00f3loga molecular de la Universidad de Auckland (Australia) opina &nbsp;\u201cEste es un avance fascinante que podr\u00eda solucionar algunos de los problemas actuales de la edici\u00f3n gen\u00e9tica, aunque a\u00fan queda tiempo hasta demostrar que puede corregir errores en el tipo de c\u00e9lulas esperado y en contextos cl\u00ednicos\u201d,<\/p>\n\n\n\n<p>El investigador en Biolog\u00eda Sint\u00e9tica de la Universitat Pompeu Fabra (UPF), Marc G\u00fcell, que tambi\u00e9n conf\u00eda en que los resultados \u00abde este grand\u00edsimo trabajo\u00bb.
